I have only had a request for one such letter in my entire lifetime. The prospective Tenant was really unable to take possession. Seems he was in the joint. I think he may have been building a collection of documentation to produce in his upcoming trial. Go figure.
Or the tenant is hoping that in your letter you will admit to not renting to him on some basis that is illegal. Remember it is illegal to discriminate in the provision of housing on the basis of race, religion, age, source of income and one or two other things I've forgotten since I don't do residential any more.
If you're sending a denial letter make sure you don't turn them down for any of these reasons.

Is it important to you that you have proof of delivery?
Its not even necessary to send them a letter. Why do you want to do so?
Waste of time and postage. If they do not recieve your call they will come to the right conclusion
