Send out enough to get a statistically significant sample. Measure the response rate and the types of responses.
WHen you change something change track what was changed and run an actual test (half the letters are the only one with the known response rate. Half are the new letter).
Which ever letter produces the best results becomes the new control letter. Then you look at more tweaks to see if you can make improvements once again.
